Nijmegen breakage syndrome (NBS) is a rare, recessively inherited cancer predisposition disorder characterized by microcephaly, immunodeficiency, and predisposition to leukemia and solid tumors. The affected genes include a 60 kDa subunit of the Mre11-Rad50-Nbs1 (MRN) complex and a protein known as NBS1. NBS1, a nuclear phosphoprotein, contains a 3'-untranslated region (3'UTR) in the form of a short and long mRNA. Short and long 3'UTRs have different lengths of poly(A) tails, and have been reported to exist as A and B isoforms. We report here that both A and B mRNA isoforms exist in multiple types of human cells. In A form-expressing cells, both the long and short 3'UTR exist, while in B form-expressing cells, only the long 3'UTR exists. The size of 3'UTR-long poly(A) tail was observed to change in the presence of RNase H, a component of the ribonucleoprotein complex, suggesting that the 3'UTR of B form mRNA is directly linked to poly(A) tails. However, the elongation of 3'UTR-long poly(A) tail was not observed in the presence of protein synthesis inhibitor. We propose that the length of 3'UTR-long poly(A) tail changes in the presence of protein synthesis inhibitor, which induces the conformational change in the long form of NBS1 mRNA.
